Description: According
to enzyme substrate method, mold and yeast chromogenic medium distinguish
between different molds and yeasts by color, after culture for 3-5 days at
25-28 ℃, the colonies of Candida albicans are
blue-green, Candida tropicalis are blue-green, Candida glabrata are blue-green,
Candida krusei are blue-green, Saccharomyces cerevisiae are blue-green,
blue-green mold are cotton-like blue-green, bacteria are suppressed.

Instructions:
Suspend 9.5g/200mL (according to the
proportion of 4.75g / 100mL to adjust), boil in one minute. After cooling to
45-50 ℃in a water bath, gently shake and pour into
sterile petri dishes (15ml for 9cm dish, 10ml for 7cm dish), after agar
completely solidification, inverted culture in incubator at 36 ℃
± 1 ℃ for 0.5-1.
Procedures:
1. Prepared sample solution according to
national standards, SN standard, FDA standards or other methods.
2. Take sample solution, coat or streaked
on the plates
3. culture for 3-5 days at 25-28 ℃, the colonies of Candida albicans are blue-green,
Candida tropicalis are blue-green, Candida glabrata are blue-green, Candida
krusei are blue-green, Saccharomyces cerevisiae are blue-green, blue-green mold
are cotton-like blue-green, bacteria are suppressed.
4. For undermined colonies of Candida
albicans can be streaked onto PDA agar plates, 25-28 ℃ for 24-48 hours, select single colony to do
microscopy and biochemical tests
Quality Control:
1. Appearance:
The dehydrate powder medium is homogeneous,
having good flowability, milky white, prepared medium is colorless solid.
2. Microbial sensitivity test:
Incubated at 36 ± 1 ℃ 18-24h:

Methods limitations:
The medium identification of mold and yeast,
may occur false-positive in a few cases, but no false-negative, sometimes it needs
to do other complementary tests
Typical features:
The colonies of Candida albicans are
blue-green, Candida tropicalis are blue-green, Candida glabrata are blue-green,
Candida krusei are blue-green, Saccharomyces cerevisiae are blue-green,
blue-green mold are cotton-like blue-green, bacteria are suppressed.
Storage:
The prepared plates must be used
immediately, avoid direct exposure to light. The powder medium should be placed
in a dark, dry place, storage temperature 2-8 ℃, avoid direct exposure to light.
Failure:
Dry powder media which are over the shelf
life, caking and color variation cannot be used.
Note:
This medium is only for laboratory use.
